Strength
  • Tramadol 100 mg mg
  • Domperidone 20 mg
Packing Size
  • 10 Tablets

Mechanism

Tramadol: Centrally acting synthetic opioid → binds to μ-opioid receptors and inhibits reuptake of norepinephrine and serotonin → analgesic effect.
Domperidone: Dopamine D2 receptor antagonist → enhances gastric motility and prevents nausea/vomiting.

Pharmacokinetics

  • Absorption: Both rapidly absorbed orally; Tramadol peaks in 1–2 hrs; Domperidone in ~30 min.
  • Distribution: Tramadol widely distributed, ~20% protein bound; Domperidone ~91–93% protein bound.
  • Metabolism: Tramadol metabolized hepatically via CYP2D6 & CYP3A4; Domperidone via CYP3A4.
  • Excretion: Both primarily excreted via urine and feces.
  • Half-life: Tramadol ~6 hrs; Domperidone ~7 hrs.

Side Effects

  • Nausea, vomiting (Tramadol)
  • Dizziness, drowsiness
  • Constipation
  • Risk of dependence (Tramadol)
  • QT prolongation (Domperidone, rare)

Contraindication

  • Hypersensitivity to Tramadol or Domperidone
  • Severe hepatic impairment
  • History of opioid dependence
  • Cardiac arrhythmias (Domperidone)

Interaction

  • Alcohol increases hepatotoxicity risk (Tramadol)
  • Tramadol may interact with SSRIs, MAO inhibitors (↑ serotonin syndrome risk)
  • Domperidone may interact with QT-prolonging drugs
  • Other CNS depressants increase sedation

Warning and Precautions

  • Do not exceed recommended daily dose
  • Monitor liver function during prolonged use
  • Use cautiously in patients with seizure disorders
  • Monitor cardiac function with Domperidone

Dosage

As directed by the Physician.
