Rabeprazole + Domperidone

Tablet

This combination provides dual action: Rabeprazole reduces gastric acid secretion, while Domperidone enhances gastric motility and prevents nausea. It is prescribed for GERD, acid reflux, dyspepsia, and nausea associated with delayed gastric emptying.

Speciality Gastroenterology | Acid Suppression | Prokinetic Therapy
Tags GERD | Acid Reflux | Dyspepsia | Nausea Relief | Proton Pump Inhibitor | Prokinetic
Strength
  • Rabeprazole 20 mg/40 mg
  • Domperidone 10 mg
Packing Size
  • 10 Tablets

Mechanism

Rabeprazole: Proton pump inhibitor → irreversibly blocks H+/K+ ATPase in gastric parietal cells → reduces gastric acid secretion.
Domperidone: Dopamine D2 receptor antagonist → enhances gastric emptying and motility → reduces nausea and vomiting.

Pharmacokinetics

  • Absorption: Rabeprazole rapidly absorbed, peak plasma levels in 3–4 hrs; Domperidone absorbed orally with peak in ~30 min.
  • Distribution: Rabeprazole ~96% protein bound; Domperidone ~91–93% protein bound.
  • Metabolism: Rabeprazole metabolized hepatically via CYP2C19 & CYP3A4; Domperidone via CYP3A4.
  • Excretion: Rabeprazole excreted via urine and feces; Domperidone primarily via feces.
  • Half-life: Rabeprazole ~1–2 hrs; Domperidone ~7 hrs.

Side Effects

  • Headache
  • Diarrhea or constipation
  • Abdominal pain
  • Dry mouth
  • Rare: QT prolongation (Domperidone)

Contraindication

  • Hypersensitivity to Rabeprazole or Domperidone
  • Severe hepatic impairment
  • Cardiac arrhythmias (Domperidone)

Interaction

  • Rabeprazole may reduce absorption of drugs requiring acidic pH (e.g., ketoconazole)
  • Domperidone may interact with QT-prolonging drugs
  • Concurrent use with strong CYP3A4 inhibitors increases Domperidone levels

Warning and Precautions

  • Do not exceed recommended daily dose
  • Monitor liver function during prolonged use
  • Use cautiously in patients with cardiovascular disorders
