Paracetamol + Domperidone

Tablet

Paracetamol + Domperidone is a fixed-dose combination used for the management of fever, pain, and associated nausea or vomiting. Paracetamol provides antipyretic and analgesic action, while Domperidone enhances gastric motility and prevents vomiting. Together, they offer comprehensive relief in febrile and painful conditions with GI symptoms.

Strength
  • Paracetamol 325 mg
  • Domperidone 10 mg
Packing Size
  • 10 Tablets

Mechanism

Paracetamol: Inhibits prostaglandin synthesis in the CNS, reducing fever and pain.
Domperidone: Dopamine (D2) receptor antagonist that enhances gastric emptying and provides antiemetic effect. The combination ensures fever and pain relief with control of nausea and vomiting.

Pharmacodynamics:

Like other 2nd generation antihistamines, loratadine is selective for peripheral H1 receptors.Loratadine does not penetrate effectively into the central nervous system and has poor affinity for CNS H1-receptors.These qualities result in a lack of CNS depressant effects such as drowsiness, sedation, and impaired psychomotor function. The onset of action varies significantly and clinical efficacy is not always directly related to only the H1 receptor potency, as concentration of free drug at the receptor must also be considered.

Pharmacokinetics

  • Absorption: Paracetamol rapidly absorbed; Domperidone bioavailability ~15%.
  • Distribution: Both widely distributed; Domperidone protein binding ~90–95%.
  • Metabolism: Paracetamol via hepatic conjugation; Domperidone via CYP3A4.
  • Elimination: Paracetamol in urine; Domperidone in urine and feces.
  • Half-life: Paracetamol ~2–3 hrs; Domperidone ~7–9 hrs.

Side Effects

  • Headache
  • Dizziness
  • Abdominal discomfort
  • Dry mouth
  • Diarrhea or constipation
  • Rare: QT prolongation (Domperidone)
  • Rare: Liver toxicity (high-dose Paracetamol)

Contraindication

  • Hypersensitivity to Paracetamol or Domperidone
  • Severe hepatic impairment
  • Cardiac conduction abnormalities (QT prolongation)
  • Concomitant use with potent CYP3A4 inhibitors

Interaction

  • Alcohol (increases liver toxicity risk with Paracetamol)
  • Ketoconazole, erythromycin (increase Domperidone levels)
  • Other QT-prolonging drugs
  • Warfarin (Paracetamol may alter INR)

Warning and Precautions

  • Use under medical supervision only
  • Do not exceed recommended Paracetamol dose (risk of hepatotoxicity)
  • Caution in elderly and patients with cardiac risk
  • Consult physician during pregnancy and lactation

Dosage

As directed by the Physician.
