Omeprazole + Domperidone

Tablet

Omeprazole + Domperidone is a fixed-dose combination used in the management of acid-related gastrointestinal disorders. Omeprazole reduces gastric acid secretion, while Domperidone enhances gastric motility and provides antiemetic action. Together, they provide effective relief from GERD, dyspepsia, and associated nausea.

Strength
  • Omeprazole 20 mg/40 mg
  • Domperidone 10 mg
Packing Size
  • 10 Tablets

Mechanism

Omeprazole: Proton pump inhibitor that irreversibly blocks H+/K+ ATPase in gastric parietal cells, reducing acid secretion.
Domperidone: Dopamine (D2) receptor antagonist that enhances gastric emptying and provides antiemetic effect. The combination ensures acid suppression and improved gastric motility.

Pharmacodynamics:

Like other 2nd generation antihistamines, loratadine is selective for peripheral H1 receptors.Loratadine does not penetrate effectively into the central nervous system and has poor affinity for CNS H1-receptors.These qualities result in a lack of CNS depressant effects such as drowsiness, sedation, and impaired psychomotor function. The onset of action varies significantly and clinical efficacy is not always directly related to only the H1 receptor potency, as concentration of free drug at the receptor must also be considered.

Pharmacokinetics

  • Absorption: Omeprazole rapidly absorbed; Domperidone bioavailability ~15%.
  • Distribution: Both widely distributed; Domperidone protein binding ~90–95%.
  • Metabolism: Omeprazole via CYP2C19 & CYP3A4; Domperidone via CYP3A4.
  • Elimination: Omeprazole in urine; Domperidone in urine and feces.
  • Half-life: Omeprazole ~1 hr; Domperidone ~7–9 hrs.
