Mechanism

Omega-3 Fatty Acids (EPA & DHA):

  • EPA (Eicosapentaenoic Acid) and DHA (Docosahexaenoic Acid) are incorporated into phospholipid membranes, altering membrane fluidity and receptor function.
  • They reduce synthesis of pro-inflammatory eicosanoids (like prostaglandin E2 and leukotriene B4) and promote anti-inflammatory mediators (resolvins, protectins).
  • EPA lowers triglyceride levels by reducing hepatic VLDL synthesis and enhancing β-oxidation of fatty acids.
  • DHA supports neuronal membrane integrity, synaptic plasticity, and neurotransmitter release, improving cognitive and visual functions.
  • Both improve endothelial function, reduce platelet aggregation, and enhance blood flow, lowering cardiovascular risk.

Lycopene:

  • A carotenoid with strong singlet oxygen quenching ability, neutralizing free radicals and reducing oxidative stress.
  • Modulates gene expression related to cell cycle regulation, apoptosis, and antioxidant defense.
  • Inhibits LDL oxidation, reducing atherosclerotic plaque formation.
  • Protects DNA from oxidative damage, lowering risk of mutagenesis and carcinogenesis.
  • Supports prostate health by downregulating IGF-1 signaling and reducing inflammatory cytokines.

Selenium:

  • Essential trace element incorporated into selenoproteins such as glutathione peroxidase and thioredoxin reductase.
  • These enzymes catalyze reduction of hydrogen peroxide and lipid hydroperoxides, protecting cells from oxidative damage.
  • Selenium regenerates Vitamin E and Vitamin C, enhancing the antioxidant network.
  • Supports thyroid hormone metabolism via iodothyronine deiodinases, regulating energy balance and growth.
  • Modulates immune response by enhancing T-cell proliferation and antibody production.

Pharmacokinetics

  • Absorption: Omega-3 fatty acids are absorbed in the small intestine; Lycopene absorption enhanced with dietary fat; Selenium absorbed efficiently.
  • Distribution: EPA & DHA incorporated into cell membranes, especially in brain, retina, and cardiovascular tissues.
  • Metabolism: Omega-3s metabolized in the liver via β-oxidation; Lycopene metabolized into various oxidative derivatives; Selenium incorporated into selenoproteins.
  • Elimination: Omega-3s excreted via urine and bile; Lycopene via feces; Selenium primarily via urine.
  • Half-life: EPA/DHA ~2–3 days; Lycopene ~2–3 days; Selenium ~1 day depending on status.

Side Effects

  • Nausea, diarrhea, abdominal pain
  • Taste perversion, rash, elevated ALT, increased LDL-C (long-term use)
