Mechanism

Modafinil enhances wakefulness by influencing neurotransmitters such as dopamine, norepinephrine, and histamine. It increases hypothalamic activation and reduces sleep drive, promoting alertness without significant cardiovascular stimulation.

Pharmacokinetics

  • Absorption: Rapid oral absorption; peak plasma concentration in 2–4 hours.
  • Distribution: Widely distributed; ~60% protein bound.
  • Metabolism: Hepatic metabolism via CYP3A4, CYP2C19, and CYP2C9.
  • Elimination: Renal and hepatic excretion.
  • Half-life: Approximately 12–15 hours.
