Mechanism

Methimazole inhibits thyroid peroxidase enzyme, blocking the iodination of tyrosine residues and coupling reactions in thyroglobulin. This prevents synthesis of thyroid hormones (T3 and T4).

Pharmacokinetics

  • Absorption: Calcium absorbed in the small intestine (requires vitamin D); magnesium absorbed in ileum and colon; zinc absorbed in jejunum.
  • Distribution: Calcium stored in bones and teeth; magnesium distributed in muscle and soft tissues; zinc bound to albumin and widely distributed.
  • Metabolism: Minerals are not metabolized but act as cofactors in enzymatic reactions.
  • Elimination: Calcium excreted via feces and urine; magnesium primarily via urine; zinc via feces and urine.
  • Half-life: Calcium and magnesium levels depend on dietary intake and bone turnover; zinc turnover is relatively rapid with daily requirement replenishment.
