Lorazepam

Tablet

Lorazepam is a benzodiazepine used for short-term management of anxiety, insomnia, seizures, and sedation. It enhances the effect of GABA at the GABA-A receptor, producing calming, muscle-relaxant, and anticonvulsant effects. Due to its high risk of dependence, it is recommended only for limited duration under strict medical supervision.

Mechanism

Lorazepam enhances GABAergic neurotransmission at the GABA-A receptor, increasing chloride ion influx and neuronal hyperpolarization. This produces anxiolytic, sedative, hypnotic, muscle relaxant, and anticonvulsant effects.

  • Absorption: Rapidly absorbed orally.
  • Distribution: Widely distributed, crosses blood-brain barrier.
  • Metabolism: Hepatic metabolism (glucuronidation).
  • Elimination: Excreted mainly via urine as metabolites.
