Glucosamine + Diacerein

Tablet

Diacerein is an anthraquinone derivative with anti-inflammatory properties that inhibits interleukin-1 activity, reducing cartilage degradation. Glucosamine is a naturally occurring compound that supports cartilage synthesis and elasticity. Together, they provide synergistic benefits in osteoarthritis management by reducing pain, slowing disease progression, and improving joint function.

Mechanism

Diacerein inhibits interleukin-1 mediated cartilage degradation and reduces inflammation. Glucosamine supports synthesis of glycosaminoglycans and proteoglycans, essential for cartilage structure and elasticity. The combination helps relieve symptoms and slow progression of osteoarthritis.

Pharmacokinetics

  • Diacerein Absorption: Well absorbed orally, metabolized to rhein
  • Glucosamine Absorption: Rapid oral absorption, concentrated in joint tissues
  • Distribution: Both widely distributed in synovial fluid and cartilage
  • Metabolism: Diacerein hepatic metabolism; Glucosamine minimal hepatic metabolism
  • Elimination: Diacerein via renal and fecal routes; Glucosamine primarily renal
  • Half-life: Diacerein ≈ 4–6 hours; Glucosamine ≈ 2–3 hours

Side Effects

  • Diarrhea (common with Diacerein)
  • Abdominal discomfort
  • Nausea
  • Skin rash (rare)

Contraindication

  • Hypersensitivity to Diacerein or Glucosamine
  • Severe hepatic or renal impairment
  • Shellfish allergy (Glucosamine source)

Interaction

  • Diacerein: May interact with laxatives (enhanced GI effects)
  • Glucosamine: May increase bleeding risk with warfarin and other anticoagulants
  • Use cautiously with hepatotoxic drugs (monitor liver function)
  • Consult healthcare provider before combining with other joint supplements

Warning and Precautions

  • Monitor liver function during therapy
  • Use cautiously in elderly patients
  • Adjust dose in case of persistent diarrhea
