Mechanism

Famciclovir is converted to penciclovir in the body. Penciclovir inhibits viral DNA polymerase, blocking viral DNA synthesis and replication. This reduces viral load and accelerates healing of lesions.

Pharmacokinetics

  • Absorption: Rapid oral absorption; bioavailability ~77%.
  • Distribution: Widely distributed in tissues.
  • Metabolism: Converted to active penciclovir via hepatic metabolism.
  • Elimination: Excreted primarily in urine.
  • Half-life: ~2–3 hours (penciclovir intracellular half-life longer).

Side Effects

  • Headache
  • Nausea
  • Diarrhea
  • Dizziness
  • Fatigue

Contraindication

  • Hypersensitivity to Famciclovir or Penciclovir
  • Severe renal impairment (dose adjustment required)

Interaction

  • Probenecid: May increase penciclovir plasma levels.
  • Other nephrotoxic drugs: Increased risk of renal toxicity.

Warning and Precautions

  • Adjust dose in patients with renal impairment.
  • Use cautiously in elderly patients.
  • Not a cure for herpes — reduces symptoms and recurrence.
  • Advise patients to avoid sexual contact during active lesions.

Dosage

As directed by the Physician.
