Doxylamine, Pyridoxine & Folic Acide

Tablet

This fixed-dose combination is widely prescribed for the management of nausea and vomiting during pregnancy. Doxylamine Succinate, an antihistamine, provides antiemetic effects. Pyridoxine Hydrochloride (Vitamin B6) reduces nausea, while Folic Acid supports fetal neural development. Together, they provide safe and effective relief for expectant mothers.

Mechanism

Doxylamine Succinate acts as an H1 receptor antagonist, reducing nausea and vomiting. Pyridoxine Hydrochloride (Vitamin B6) modulates neurotransmitter synthesis, alleviating nausea. Folic Acid supports DNA synthesis and neural tube development in the fetus.

Pharmacokinetics

  • Doxylamine: Rapid absorption, hepatic metabolism, half-life ≈ 10 hours
  • Pyridoxine: Well absorbed, converted to active pyridoxal phosphate, half-life ≈ 15–20 days
  • Folic Acid: Absorbed in small intestine, converted to tetrahydrofolate, half-life ≈ 3–4 hours

Side Effects

  • Drowsiness
  • Dry mouth
  • Dizziness
  • Rare: allergic reactions

Contraindication

  • Hypersensitivity to any component
  • Severe hepatic impairment

Interaction

  • Alcohol and CNS depressants — additive sedation with Doxylamine
  • Levodopa — reduced efficacy with Pyridoxine
  • Antiepileptics — may reduce folate levels

Warning and Precautions

  • Use under medical supervision during pregnancy
  • Monitor for excessive sedation
  • Ensure adequate folate supplementation
