Metoprolol + Amlodipine

Tablet

Tablet contains a combination of two medicines; Amlodipine and metoprolol which help to control high blood pressure. Lowering blood pressure helps to reduce your risk of having a heart attack or a stroke in the future. Amlodipine is a calcium channel blocker which inhibit the entry of calcium ion and leads to dilation (widens) of blood vessels and improves blood flow. Amlodipine is indicated for hypertension. Lowering blood pressure prevents cardiovascular diseases such as stroke or heart attack. Metoprolol is a selective beta-1 blocker which slows down the conduction of heart and decreases the blood pressure gradually.

Strength
  • Metoprolol 50 mg
  • Amlodipine 5 mg
Packing Size
  • 10 Tablets

Mechanism

Metoprolol Succinate: Selectively blocks beta-1 adrenergic receptors, reducing heart rate, cardiac output, and myocardial oxygen demand.
Amlodipine Besylate: Inhibits calcium influx into vascular smooth muscle, causing vasodilation and lowering blood pressure. The combination ensures effective blood pressure control and angina relief.

Pharmacokinetics

  • Absorption: Metoprolol well absorbed; Amlodipine bioavailability ~60–65%.
  • Distribution: Both widely distributed; Metoprolol protein binding ~12%; Amlodipine ~93%.
  • Metabolism: Metoprolol via CYP2D6; Amlodipine via CYP3A4.
  • Elimination: Metoprolol renal; Amlodipine renal and fecal.
  • Half-life: Metoprolol Succinate ~3–7 hrs (prolonged release extends effect); Amlodipine ~30–50 hrs.

Side Effects

  • Dizziness
  • Fatigue
  • Bradycardia (Metoprolol)
  • Peripheral edema (Amlodipine)
  • Headache

Contraindication

  • Hypersensitivity to Metoprolol or Amlodipine
  • Severe bradycardia or heart block
  • Cardiogenic shock
  • Severe hypotension

Interaction

  • Other antihypertensives (additive effect)
  • CYP3A4 inhibitors (increase Amlodipine levels)
  • CYP2D6 inhibitors (increase Metoprolol levels)
  • NSAIDs (may reduce antihypertensive effect)

Warning and Precautions

  • Use under medical supervision only
  • Monitor blood pressure and heart rate regularly
  • Caution in hepatic impairment
  • Consult physician during pregnancy and lactation
