Fosfomycin Tromethamine

SACHET

Fosfomycin Tromethamine is an oral antibacterial medicine that is converted to active fosfomycin after administration. It acts by inhibiting an early step in bacterial cell-wall synthesis. The oral granule formulation is used for appropriately susceptible bacterial infections, with current prescribing information specifically indicating it for acute uncomplicated cystitis in adult women caused by susceptible strains of Escherichia coli and Enterococcus faecalis. It is not indicated for pyelonephritis or perinephric abscess in that prescribing information.

Mechanism of Action

Fosfomycin is a bactericidal antibacterial agent that inhibits an early and essential step in bacterial cell-wall synthesis. It is transported into susceptible bacterial cells through specific uptake systems and then binds covalently to UDP-N-acetylglucosamine enolpyruvyl transferase (MurA). This enzyme is required for the initial stage of peptidoglycan synthesis, an essential component of the bacterial cell wall. By inhibiting MurA, fosfomycin blocks peptidoglycan formation, resulting in disruption of bacterial cell-wall synthesis and subsequent bacterial cell death.

Indications

Fosfomycin Tromethamine is indicated for the treatment of acute uncomplicated cystitis in adult women caused by susceptible bacterial strains, according to the approved prescribing information.
