Amantadine is an antiviral and antiparkinsonian agent. It is used in the management of Parkinson’s disease and drug-induced extrapyramidal symptoms due to its dopaminergic activity. It also has activity against influenza A virus, though resistance limits its use.

Mechanism

Amantadine increases dopamine release and inhibits dopamine reuptake in the brain, improving motor symptoms in Parkinson’s disease. As an antiviral, it blocks the M2 ion channel of influenza A virus, preventing viral replication.

Absorption: Well absorbed orally.
Distribution: Widely distributed; crosses blood-brain barrier.
Metabolism: Minimal hepatic metabolism.
Elimination: Excreted unchanged in urine.
Half-life: Approximately 10–15 hours.
